L'étude historique de l'ostréiculture française monte que cette activité est caractérisée par des périodes de prospérités et de pénurie. Des épizooties ont été à l'origine de ces alternances. A chacune de ces période, on a pu constater des changements d'espèces : Ostrea edulis fut remplacée par Crassostrea angulata, de même celle-ci fut remplacée par Crassostrea gigas dans les années 1970. L'étude de la bibliographie montre que les techniques utilisées pour distinguer des espèces Crassostrea angulata et Crassostrea gigas sont insuffisantes. Parmi les maladies infectieuses observées chez les mollusques bivalves marins, les viroses sont souvent mal connues, en raison d'une certaine inadéquation des techniques d'identification et de diagnostic généralement mises en oeuvre lors de phénomènes de mortalité. En effet, la microscopie photonique a été et reste encore, dans de nombreux laboratoires travaillant sur les pathologies des mollusques, la technique de base pour l'analyse des échantillons. Les parasites du genre Marteilia sont responsables de graves épizooties et sont retrouvés dans une communauté très variable de mollusques bivalves marins. Toutefois ces protistes restent difficiles à classer à l'intérieur du genre du fait que les données morphologiques sont insuffisantes pour distinguer avec précision deux espèces données. Ceci est le cas pour Martilia refringens, affectant l'huître plate Ostrea edulis ou la moule (Mytilus edulis et Mytilus galloprovincialis), Marteilia sp. et Marteilia maurini affectant les deux espèces de moules. A set of primers were designed according to the published sequence of the gene encoding a rhoptry protein of Babesia caballi, and used to amplify parasite DNA from the blood samples obtained from carrier horses by polymerase chain reaction(PCR)method.The PCR method was sensitive enough to detect parasite DNA from 2.5 μl blood sample with a parasitemia of 0.000001%. The PCR method was compared with fluorescent antibody test(IFAT) in order to evaluate the diagnosis effciency for B. caball infection in horses. Of 142 field samples from Mongolia, 28(20%) and 96(69%)samples were identified positively by PCR and IFAT, respectively. Seed pathology as a subdisipline of plant pathology is relatively new. Paul Neergard is considered the father of seed pathology. Recent developments in the area of seed pathology technology allow for more ecofriendly seed treatments and more reliable seed health testing. Due to economics and new interest in environmental issues, research into the viability of biological seed treatments is becoming more common. The use of sophisticated DNA amplification technologies allows for the detection of seedborne pathogens that might go undetected using more conventional means. These types of research will be fundamental in guaranteeing seed health quality standards and achieving phytosanitary requirements throughout the world in the new millennium. |

Avec 30 000 tonnes d'huîtres produites chaque année, l'ostréiculture est l'activité principale du bassin de Marennes-Oléron. Caractérisé par une très forte interaction avec les milieux professionnels de la conchyliculture, l'Ifremer se doit avant tout de répondre à leurs questions, en particulier en matière de croissance, de reproduction et surtout en matière de mortalité de l'huître creuse. En effet, l'huître portugaise Crassostrea angulata fut décimée de 1968 à 1972 par un agent pathogène de type iridovirus. Mycobacterium avium complex (MAC) species cannot be discriminated by the usual methods of biochemical identification of mycobacteria. This study showed that amplification by PCR of DT1 and DT6, two single copy sequences identified in the genome of M. avium serotype 2, the insertion sequence IS1245, found to be consistently present in M. avium strains and the heat-shock protein gene hsp65, followed by restriction polymorphism analysis, are rapid and accurate tests for the differentiation of the species M. avium, M. intracellulare, and M. scrofulaceum. |

Informações sobre peso à desmama de um rebanho Nelore foram utilizadas após ajuste para idade padrão de 205 dias, sexo da cria, idade da mãe, touro e mês de desmama, para separar as reprodutrizes em dois grupos, cujos filhos diferiam nesse peso. As médias ajustadas pelo método dos quadrados mínimos foram para os grupos pesado (P) e leve (L) de 163,21± 2,18kg e 134,44± 2,18kg, respectivamente, com 41 animais em cada grupo. Essas reprodutrizes foram submetidas à coleta de sangue para estudo de polimorfismos do gene da somatotropina bovina, pela técnica de PCR-RFLP (polymerase chain reaction-restriction fragment length polymorphism). We have developed a procedure for nonradioactive single strand conformation polymorphism analysis and applied it to the detection of point mutations in the human tumor suppressor gene p53. The protocol does not require any particular facilities or equipment, such as radioactive handling, large gel units for sequencing, or a semiautomated electrophoresis system. This technique consists of amplification of DNA fragments by PCR with specific oligonucleotide primers, denaturation, and electrophoresis on small neutral polyacrylamide gels, followed by silver staining. The sensitivity of this procedure is comparable to other described techniques and the method is easy to perform and applicable to a variety of tissue specimens. |
